Abstract Text: Prnp is expressed in most cell types and suspected that its expression is tightly regulated during. It has been shown that prion diseases show sex-specific expression, but tissue-specific or hormone-specific regulation has not been revealed to a large extent. Therefore, in order to understand the regulation of Prnp expression during estrous cycle, we analyzed the expression pattern of the Prnp gene in Estrogen target organs, the ovary and uterus, using various models. We analyzed the expression profile and methylation in the CpG island and promotor regions. Employing ovariectomized (OVX) mice model was used to characterize the expression of Prnp mRNA. The expression levels were different by the stages of the estrous cycle both in uterus and ovary. Its expression was higher at estrous stage than other stages. In OVX model, Prnp mRNA levels were increased by estrogen administration. The methylation patterns were also different by the stage of estrous cycle. Based on these results, it is suggested that Prnp expression in ovary and uterus is under the control of steroid hormones and epigenetic modification.
